MSCs desensitize CRPC cells to docetaxel in vivo. a MSCs were transfected with the adenoviral vector GFP-mock (Invitrogen) to be marked. After transfection about 48 h, MSCs-GFP were detected by fluorescence microscope (original magnification ×200). b Mice with PC3 tumors were injected with MSCs-GFP or not through tail vein every 3 days, while mice were treated with docetaxel (DTX) or not. Tumor volume was observed and calculated using the formula: volume = width2 × length × 0.5236. c After docetaxel (DTX) injection for 15 days, tumor tissues were removed from mice (tumors from untreated MSCs-GFP mice as control) for the further experiments. Tumor weights were measured. d Tumor tissues were embedded in Tissue-Tek OCT compound and snap frozen in liquid nitrogen. Cryostat sections (6 mm thick) were prepared using a Leica CM1950 cryostat. GFP fluorescence signal was then analyzed with a fluorescence microscope (original magnification ×200). *P < 0.05; **P < 0.01
