From: Dimerized translationally controlled tumor protein increases interleukin-8 expression through MAPK and NF-κB pathways in a human bronchial epithelial cell line

Fig. 1

IL-8 induction by dTCTP is transcriptionally regulated. a BEAS-2B cells were incubated with 10 μg/ml of dTCTP for 6 or 24 h, and expression of IL-8 mRNA and β-actin mRNA was determined by RT-PCR. Data shown are representative of three independent experiments. b BEAS-2B cells were treated with indicated concentrations of dTCTP for 6 h. Levels of IL-8 mRNAs were measured using TaqMan-based-real time PCR, normalized to levels of GAPDH mRNA, and expressed as fold increases over control. Values are expressed as fold changes (mean ± SEM, n = 3). *p < 0.05, **p < 0.01 and ***p < 0.001, significantly different from untreated group. c BEAS-2B cells were pretreated for 1 h with 0.1–1 μg/ml of actinomycin D, and coincubated with 10 μg/ml dTCTP for an additional 24 h, followed by assay for IL-8 of the supernatants. Data shown are the mean with SEM of three independent experiments. ***p < 0.001 compared with bar 1 and †††p < 0.001 compared with bar 2
